Abstract:

Background and Objectives: Improper use of chemical pesticides such as Mancozeb (MZB) leads to genital injuries. The aim of this study was to determine the effect of different doses of olive leaf extract (OLE) on reproductive damage in male mice exposed to MZB. Materials and Methods: In this experimental study, 64 adult male NMRI mice (10 to 12 weeks) were randomly divided into 8 groups (n=8) including control (Normal saline recipient), Mancozeb (MZB) recipient (500 mg/kg), MZB (500 mg/kg) + (200, 300 and 400 mg of OLE) and OLE group at doses of (200, 300 and 400 mg) and were treated by gavage every other day for 35 days. Then, the mice were killed by cervical dislocation; heart blood samples were drawn to measure serum testosterone level and epididymis removed to examine sperm. Different indicators of sperm analysis (number, motility and morphology), protamine and chromatin health were examined. Data were analyzed using one-way ANOVA and Tukey’s multiple comparisons test. Results: The lowest protamine levels (p=0.001) and the lowest alive sperms (p=0.005) were seen in the MZB group. There was a significant decrease in head abnormalities (p=0.003) and immobile sperm (p< 0.001) in OLE groups. At a dose of 200 mg/kg OLE, the highest chromatin health (p<0.001), the highest rapid sperm motility (p=0.037) and the highest testosterone level (p=0.001) were observed. Conclusion: The highest efficacy of OLE was seen at a dose of 200 mg/kg; therefore, consumption of this extract in the same dose can probably be considered as a supportive compound in the diet of high-risk people exposed to MZB. Key words: Mancozeb, Sperm, Olive leaf extract, Mice, Infertility   Funding: This study was funded by Shiraz Branch of Islamic Azad University. Conflict of interest: None declared. Ethical approval: The Ethics Committee of Shiraz Branch of Islamic Azad University approved the study (IR.IAU.SHIRAZ.REC.1399.027).   How to cite this article: Ashkanani M, Akbari H. Protective Effects of Curcumin on Sperm and Stereological Parameters in Testes of Formaldehyde-Exposed NMRI Mice: An Experimental Study

Background and Aims: Formaldehyde (FA) exposure is an important cause of cellular injury and oxidative damage in testis, leading to infertility. This study aimed to assess the protective effects of curcumin on sperm and stereological parameters in testes from formaldehyde-exposed NMRI mice. Materials and Methods: At 6-8 weeks of age, 24 adult male NMRI mice weighing 30-35 g were categorized in...

P-61: Effects of Acrylamid on Sperm Parameters and Chromatin Quality in Mice

Background: Acrylamid (AA) is found in carbohydraterich foods that are prepared at high temperatures, such as French fries and potato chips. The toxic effects of AA in male animals include degeneration of epithelial cells of seminiferous tubules, decreased number of sperm and elevation in abnormal spermatozoa. The aim of this study was to evaluate the detrimental effects of AA on sperm paramete...

The Effects of Olive Leaf Extract on The Testis, Sperm Quality and Testicular Germ Cell Apoptosis in Male Rats Exposed to Busulfan

Objective Busulfan (BU) has a destructive effect on the male reproductive system. On the one hand, traditional herbs such as olive leaf extract (OLE) have been considered as sources of antioxidants and phenolic compounds. The goal of this study was to assess the effects of OLE on BU-induced damages in rat testes. MaterialsAndMethods In this experimental study, 40 male Wistar rats divided random...

P-1: Morphine Has Detrimental Effects on Sperm Parameters, Chromatin Quality and DNA Integrity in Mice

Background Morphine as a natural alkaloid (opiate) is considered as one of the problems associated with poor semen production and sperm quality. For the first time, this experimental study was carried out to evaluate the impact of intra-peritoneal injection of morphine on sperm parameters, chromatin quality and DNA integrity of spermatozoa aspirated from cauda epididymis of mice. MaterialsAndMe...

P-12: The Improvement of Sperm Parameters and Chromatin Quality by Vitamin E Supplement in Mice

Background: Vitamin E is believed to be the primary component of the antioxidant system in spermatozoa. There is a relationship between activity of these antioxidant and function of sperm. Vitamins E and C which are belong to non-enzymatic antioxidant are used as a supplemented drug to improve sperm quality and male fertility. The present study was carried out to investigate the positive effect...

Protective effect of hydroalcoholic olive leaf extract on experimental model of colitis in rat: involvement of nitrergic and opioidergic systems.

The aim of the present study is to investigate the possible protective effect of dry olive leaf extract (OLE) against acetic acid-induced ulcerative colitis in rats, as well as the probable modulatory effect of nitrergic and opioidergic systems on this protective impact. Olive leaf extract was administered (250, 500 and 750 mg/kg) orally for two successive days, starting from the colitis induct...
